Streptococcus thermophilus is widely used in food fermentations; it commonly suffers diverse stress challenges during manufacturing. This study investigated the cold shock response of S. thermophilus when the cell culture temperature shifted from 42°C to 15°C or 20°C. The growth of cells was affected more drastically after cold shock at 15°C than at 20°C. The generation time was increased by a factor of 19 when the temperature was lowered from 42° to 20°C, and by a factor of 72 after a cold shock at 15°C. The two-dimensional electrophoretic protein patterns of S. thermophilus under cold shock conditions were compared with the reference protein pattern when cells were grown at optimal temperature. Two proteins of 21.5 and 7.5 kDa synthesized in response to cold shock were characterized. N-terminal sequencing and sequence homology searches have shown that the 7.5-kDa protein belonged to the family of the major cold shock proteins, while no homology was found for the new cold shock protein of 21.5 kDa. A molecular phylogeny of the Scleractinia is reconstructed from approximately 700 nucleotides of the 5'end of the 28S rDNA obtained from 40 species. A comparison of molecular phylogenic trees with biomineralization patterns of coral septa suggests that at least five clades are corroborated by both types of data. Agaricidae and Dendrophylliidae are found to be monophyletic, that is supported by microstructural data. Conversely, Faviidae and Caryophylliidae are found to be paraphyletic: Cladocora should be excluded from the faviids, whereas Eusmilia should be excluded from the caryophylliids. The conclusion is also supported by the positions, sizes and shapes of centres of calcification. The traditional Guyniidae are diphyletic, corroborating Stolarski's hypothesis 'A'. Some results from our most parsimonious trees are not strongly statistically supported but corroborated by other molecular studies and microstructural observations. For example, in the scleractinian phylogenetic tree, there are several lines of evidence (including those from our data) to distinguish a Faviidae–Mussidae lineage and a Dendrophylliidae–Agaricidae–Poritidae–Siderastreidae lineage. From a methodological standpoint, our results suggest that co-ordinated studies creating links between biomineralization patterns and molecular phylogeny may provide an efficient working approach for a re-examination of scleractinian classification. This goal is important because in the evolutionary scheme proposed by Wells that presently remains the basic framework in coral studies, patterns of septal microstructures are involved. Validating from molecular phylogenies a given microstructural character state as a potential synapomorphy for a clade is the only way to include fossils in the coral classification, an approach that should allow the unity of coral classification to be maintained up to the origin of the phylum in the Triassic times.  相似文献

The rise and consequences of polyploidy in vertebrates, whose origin was associated with genome duplications, may be best studied in natural diploid and polyploid populations. In a diploid/tetraploid (2n/4n) geographic contact zone of Palearctic green toads in northern Kyrgyzstan, we examine 4ns and triploids (3n) of unknown genetic composition and origins. Using mitochondrial and nuclear sequence, and nuclear microsatellite markers in 84 individuals, we show that 4n (Bufo pewzowi) are allopolyploids, with a geographically proximate 2n species (B. turanensis) being their maternal ancestor and their paternal ancestor as yet unidentified. Local 3n forms arise through hybridization. Adult 3n mature males (B. turanensis mtDNA) have 2n mothers and 4n fathers, but seem distinguishable by nuclear profiles from partly aneuploid 3n tadpoles (with B. pewzowi mtDNA). These observations suggest multiple pathways to the formation of triploids in the contact zone, involving both reciprocal origins. To explain the phenomena in the system, we favor a hypothesis where 3n males (with B. turanensis mtDNA) backcross with 4n and 2n females. Together with previous studies of a separately evolved, sexually reproducing 3n lineage, these observations reveal complex reproductive interactions among toads of different ploidy levels and multiple pathways to the evolution of polyploid lineages.  相似文献

Islets of Langerhans, isolated from normal or 19-day pregnant rats, were cultured for 20 h at 37 degrees C in tissue culture medium 199. When islets were cultured in medium containing low glucose (5.5 mM), the higher adenylate cyclase activity and insulin secretory responses characteristic of islets from pregnant rats were maintained during the test period of 29 h. Islets from normal and pregnant rats were also cultured for 20 h in medium containing a very high glucose concentration (83.3 mM) in order to load the B cells with glycogen. It was found, after glycogen loading, that, while adenylate cyclase activity increased to a greater extent in islets from pregnant rats than controls, this activity was not increased in proportion to the striking changes in insulin release rate observed in pregnant rat islets. The results show that the difference in insulin secretory response between islets from normal and pregnant rats may be preserved when the islets are cultured for 20 h, and that these differences are enhanced for a variety of reasons after culture of islets in 83.3 mM glucose.  相似文献

We examined whether neuromuscular reflexes were altered with anterior loads applied to the tibio-femoral joint. A ligament testing device was modified by attaching a reflex hammer to a steel mounted frame to illicit a patellar tendon tap, while anterior directed loads displaced the tibia on the femur. Five trials were acquired while anterior-directed loads (20, 50, 100 N; counterbalanced) were applied to the posterior tibia between 20 N pre (20 N(Pre)) and post (20 N(Post)) baseline conditions on two different days. Surface electromyography (sEMG) recorded mean quadriceps (Q) and hamstring (H) reflex time (R(Time)=ms) and reflex amplitude (R(Amp)=%MVIC). A load cell on the anterior tibia measured the timing (KE(Time)=ms) and amplitude (KE(Amp)=N) of the knee extension force, and was used to calculate electromechanical delay (EMD=ms) and peak knee extension moment (KE(Mom)=Nm/kg). Data from 19 recreationally active subjects revealed good to excellent response consistency between test days and between baseline conditions for R(Time), R(Amp), KE(Time) and KE(Amp). With anterior tibial loading, R(Time) was faster at 50 N vs. 20 N(Post), and R(Amp) was greater at 20 N(Pre) vs. 20 N(Post) (Q and H) and at 50 N vs. 100 N (Q only). KE(Mom) was greater at 20 N(Pre) and 50 N vs. 20 N(Post), and EMD was shorter at 50 N vs. 20 N, 20 N(Pre) and 20 N(Post). These results suggest that knee extensor reflex responses are enhanced with low (50 N) but not moderate (100 N) anterior loading of the knee.  相似文献
